Both sit on the hill-country railway, and most routes include both. The honest difference is that Kandy is a working city with a major religious site, while Ella is a small base for walking.

VerdictChoose Kandy for temples, city energy and the start of the rail journey. Choose Ella for walks, views and a slower morning. If you only have two nights for the hills, take Ella — but do not skip the Temple of the Tooth if you can route Kandy in.
Yala vs Udawalawe
The two parks most people choose between on a south-coast trip. Yala is famous for leopards and busy; Udawalawe is open country that is more dependable for elephants and calmer.

VerdictFirst safari, or travelling with children: Udawalawe. Leopard-focused, second visit, or you are already near the east: Yala. Check current openings with the Department of Wildlife Conservation either way — closures change by zone and year.
Mirissa vs Unawatuna
Two south-coast bases an hour apart. Mirissa is a curved bay with whale trips and a bigger evening scene; Unawatuna is a sheltered swimming beach a short ride from Galle Fort.

South Coast vs East Coast
The single most useful comparison in Sri Lanka, because it is really a question about your dates. The two coasts are in season at opposite ends of the year.
